If your problem is indeed hiss and not hum or buzz, then there are two most likely sources. The first one is the tube, and you already rulled this one out.The second is a passive component. For instance, the loading resistors CAN become noisy.
One simple way to troubleshoot it is to pull out the last tube and replace it with the equivalent resistor. Say, you have a tube running with 100V on its plate, and at 5mA. Then you replace it with a 10W power resistor of 20k value, turn the unit On and listen to the noise. If it is still there - the loading resistor is the likely suspect.
This allows you to isolate the stage from the preceeding stage.
If the last stage is quiet, install its tbe back and do that to the one before the last stage.
With some practice you can isolate the source easily. But to give you more exact procedure without knowing the circuit topology would be hard.
