you can see a "silvering" at the top of the glass directly above the filament. Of course with tubes that have a top getter this isn't possible, but with tubes that have bottom (or side) getters, if you get the silvering on the top that looks like getter flashing, where there is no getter, than the tube for sure is used.
Pin condition can sometimes give a pretty good indication the tube is used, but not always.
Lastly, if the logo/writing painted on the glass wipes away incredibly easily, this is often a good sign the tube was NOT ever used (because it was never heated enough to bake on). But not always.
