The 12AU7 has a gain (mu) of 17, so in this circuit it will produce a gain of half that, or 8.5. The follower section will have a gain of mu/(mu+1) or 0.94 giving a net gain of 8. The 300B will have something like 70 volts bias, so it needs a peak voltage of 70/8 or about 9 volts peak, 6v RMS. A CD player puts out typically 2vRMS peak, most LP systems less than that. Therefore with this complement you'll need a preamp with high gain and high output voltage capability - 10 to 20dB more than usual. I tried to make this point in my earlier post but it was buried near the bottom and couched in more general terms - so I ran the numbers for this specific example.

Incidentally, the 12AU7 (and the others of that family) were made by many manufacturers with different tooling. Naturally the market is flooded with the cheapest since many more of them were made, but the really good versions can be - well - really good.
