1 ASIO and WASAPI are the tricks to bypass the Win audio.
If you use one of them, the win settings don't matter as both talk straight to the audio device.
In general WASAPI works fine with USB DACs but not with discrete sound cards.

As DACs don't understand anything about WAV, FLAC, MP3 etc. it has to be converted to LPCM. I believe it was the Squeeze box community who found out that you can improve SQ by using the server to do the decoding.
If this is valid for other devices as well I can't tell you.
If it is about using a mobile as a remote (DLNA) to control the server the whole point is irrelevant as it is local playback.
2 I'm inclined to say no. I have a preference to have the audio and server process on the same box. Scanning a large audio collection is a lot of I/O and I rather have this inside the box.
