Yes. My music computer has two USB 3 ports next to each other. One has the input from my music library which is stored on a connected HDD. The other connects to the DAC. Reverse the connections and it sounds worse. Quite a bit worse. Wny? Don't know. Oh, and in my particular case the single USB 2 port sounds a little less good than either of the USB 3 ones.
