Computer-as-source people do this all the time. One of the big advantages of using a computer as your transport (along with guaranteed accurate reads, something no real-time player can achieve, and the incredible convenience of not having to keep your music on discs) is that you can perform upsampling, change bit depths, and add DSP effects (some of which sound surprisingly good) to your bit-perfect music data.By the way, I've done quite a bit of experimenting with upsampling and haven't really found a dramatic improvement (although it probably depends on the DAC that is receiving the upsampled data...if it does its own upsampling and it is doing a poor job of it, upsampling on the computer might be better).
One thing to keep in mind is that taking 16/44 music data and turning it into 24/192 music data does not improve the data...it simply puts it in a format that *may* work better for the DAC that is processing it.
