Nope, I've never been able to hear a difference nor could I ever find a significant difference in sound on my system (see link below).

As others have said, non-contiguous data blocks on HD/SSD/memory are handled by the OS and these days, there's really no issue especially given how efficiently modern OS's handle tasks like disk buffering to reduce latency. Likewise, decompression is trivial and barely take up CPU processing. This is especially trued with FLAC which is highly efficient.

Metadata tagging data blocks in the file format is not processed on playback so will not affect anything. Unless of course you have settings like ReplayGain active.
