Go to the sound icon and right click to open "Playback devices".Select the sound device that you want to use. In this case it would probably be your USB DAC (but if it were SPDIF it would be your sound card with SPDIF output). Click the CONFIGURE button. Then hit the Test button. You should see iconic sound waves out of the speakers on the window for the duration of a short test. You should also hear sound. If not the device isn't working or your amp and speakers aren't working.
Then go back to the original Playback Devices screen. Select the device that worked and set it to be the default. You will then get a green check mark.
I assume that your amplifier and speaker are working, with the DAC selected and the volume turned up enough to hear something. But I am not going to accuse you of doing anything so foolish as to hear nothing when the amp is turned off, the wrong input is selected or the volume is turned down. I would never never be so stupid. :-)
